first time jetting

jetting the blaster this weekend. new dynoport exhaust cfm airbox elevation is 746. any suggestions on where i should start jetting? i was thinking 300 on the main. from looking at previous posts most people just switch out the mains and not the needle or pilot is this correct?
 
mine has a stock pilot with the clip in the stock position on the needle witha 290 main jet i have the factory 7 air box with factory 7 pipe and v force3 reeds
 
Yep, the pilot is optional depending on how it runs with it. Since the pilot directly controls your throttle response, I would say jet without it for the main, then throw it in and rejet if necessary. If if lessens your throttle response, then take it back out.
